ABSTRACT

Objective To analyze the efficacy and safety of pramipexole in treating restless legs syndrome (RLS) to different races.Methods A search for randomized,double-blind,and placebocontrolled clinical trials of pramipexole in treating RLS using Pubmed was carried out.A meta-analysis of included clinical trials was performed with Revman 5.2.The efficacy was calculated based on the change of the weighted mean difference (WMD) with International Restless Legs Syndrome Study Group rating scale (IRLS) and the relative risk (RR) of response with Clinical Global Impression-Improvement (CGI-I) scale.Safety was assessed with RR of the adverse event (AE).Subgroup analysis was conducted in the white race subgroup and the yellow race subgroup,separately.Results A total of 6 clinical trials were included in this meta-analysis.The records of patients were pooled.Overall,WMD of the change in IRLS score was-4.72 (Z =6.57,P < 0.01),which was-4.33 in white race subgroup (Z =3.90,P < 0.01) and-4.52 (Z =42.28,P <0.01) in yellow race subgroup.Overall RR of response based on CGI-I was 1.64 (Z =10.39,P <0.01),which was 1.60 (Z =4.66,P <0.01) in white race subgroup,and 1.51 (Z =4.42,P < 0.01) in yellow race subgroup.Overall RR for AE was 1.26 (Z =5.11,P < 0.01),which was 1.26 (Z =2.27,P =0.02) in white race subgroup and 1.36 (Z =2.98,P =0.003) in yellow race subgroup.Conclusion To treat RLS,pramipexole is an effective and safe drug.The results are similar for both the white race and the yellow race.

ABSTRACT

Antimicrobial screening of several novel 4-thiazolidinones with benzothiazole moiety has been performed. These compounds were evaluated for antimicrobial activity against a panel of bacterial and fungal strains. The strains were treated with these benzothiazole derivatives at varying concentrations, and MIC’s were calculated. Structures of these compounds have been determined by spectroscopic studies viz., FT-IR, 1H NMR, 13C NMR and elemental analysis. Significant antimicrobial activity was observed for some members of the series, and compounds viz. 3-(4-(benzo[d]thiazol-2-yl) phenyl)-2-(4-methoxyphenyl)thiazolidin-4-one and 3-(4-(benzo[d]thiazol-2-yl)phenyl)-2-(4-hydroxy phenyl)thiazolidin-4-one were found to be the most active against E.coli and C.albicans with MIC values in the range of 15.6–125 μg/ml. Preliminary study of the structure–activity relationship revealed that electron donating groups associated with thiazolidine bearing benzothiazole rings had a great effect on the antimicrobial activity of these compounds and contributes positively for the action. DNA cleavage experiments gave valuable hints with supporting evidence for describing the mechanism of action and hence showed a good correlation between their calculated MIC’s and its lethality.

ABSTRACT

Objective: The present study is to evaluate antiepileptic activity of some fluoro benzothiazolo oxadiazolo quinazoline and sulfonamido quinazoline derivatives. Background of the Study: Epilepsy is a chronic disorder of the brain, characterized by the periodic and unpredictable occurrence of seizures. Epilepsies affect around 1–2% of the world population including the fact that the convulsions of approximately 25% of epileptics are inadequately controlled by medication. Materials and Methods: Albino mice (weighing 20-25g) of either sex were used in this study. MES seizures were induced in mice by delivering electroshock of 60ma for 0.2 seconds by means of an electro- convulsiometer transauricularly through a pair of ear clip electrodes. Both test animals and standard group received diazepam (5mg/kg) p.o strychnine nitrate (1 mg/kg) was administered. Results and Discussion: MES induced convulsion the mentioned dose was administered one hour prior to MES elicitation. It was observed that all SSBDs except ap- 7, bz-10 and s-5 shown significant anticonvulsant effect. The results are compiled in the Table 1 and graphically depicted in Figs 1 and 2 respectively. Conclusion: The present investigation revealed that the SSBDS ap-3, ap-6 and bz-9 were shown significant anticonvulsant activity against both MES and strychnine induced models.

A meta-analysis of included clinical trials was performed with RevMan 5.0 software.The 2 outcomes that the weighted mean difference(WMD) of change from baseline in International RLS Study Group rating scale(IRLS) score and the relative risk (RR) of response based on the Clinical Global Impression-Improvement (CGI-I) scale score were calculated for efficacy.Safety was assessed with RR of the adverse event (AE).Results A total of 5 clinical trials were included in this meta-analysis,of which 1776 patients were randomly assigned (945 on pramipexole,831 on placebo).The records of patients were pooled.Overall WMD were - 6.34 ( Z =12.76,P < 0.01 ) for the change from baseline in IRLS score,and RR of response based on CGI-I were 1.65 (Z =10.39,P <0.01).The overall RR of pramipexole versus placebo were 1.14 ( Z =1.87,P =0.06 ) for AE.Conclusion To treat RLS,pramipexole is an effective and safe drug.
